目的观察牛黄参胶囊含药血清对人肝癌细胞HepG-2的Bax、Bcl-2的调控作用。方法建立并分为空白对照组,环磷酰胺组和牛黄参胶囊5%、10%和20%含药血清组。采用实时定量PCR检测Bax、Bcl-2 mRNA的表达水平;免疫组织化学法检测HepG-2细胞Bax、Bcl-2的表达;Western blotting检测Bcl-2蛋白的表达水平。结果牛黄参胶囊20%含药血清能在基因和蛋白水平明显上调Bax表达(P<0.01),明显抑制Bcl-2的表达(P<0.01),并具有明显的剂量依赖性。结论牛黄参胶囊含药血清能明显上调Bax表达,同时下调Bcl-2表达,可能是其诱导肝细胞凋亡的机制之一。
Objective To observe the regulation of Bax and Bcl-2 in human hepatocellular carcinoma HepG-2 cells with serum containing Niu Huang Shen capsule. Methods Establish and divided into blank control group, cyclophosphamide group and Begonia Capsule 5%, 10% and 20% drug-containing serum group. The expression of Bax and Bcl-2 mRNA was detected by real-time quantitative PCR. The expression of Bax and Bcl-2 in HepG-2 cells was detected by immunohistochemical method. The expression of Bcl-2 protein was detected by Western blotting. Results 20% serum containing Niuhuang Shen capsule could significantly up-regulate the expression of Bax at gene and protein level (P <0.01), and significantly inhibit the expression of Bcl-2 (P <0.01) in a dose-dependent manner. Conclusion Niuhuang Shen capsule containing serum can significantly up-regulate Bax expression and down-regulate the expression of Bcl-2, which may be one of the mechanisms of its induction of hepatocyte apoptosis.
