目的制备聚电解质逐层组装的壳聚糖海藻酸钠微凝胶脉冲制剂,并对制剂的结构和体外释放行为进行考察。方法使用高压静电成囊机制备海藻酸钠壳聚糖微凝胶核心,通过静电吸附原理将聚电解质逐层包裹在微凝胶表面形成包衣层。通过单因素实验考察不同物料比对聚电解质包衣微凝胶膨胀性能、释放行为的影响。结果聚电解质包衣微凝胶外观圆整,粒径分布范围窄,体外释放时滞2.67h,时滞后药物迅速释放,3h释放量为72%。结论以壳聚糖海藻酸钠为核心,聚电解质为包衣层的微凝胶可以实现脉冲释放。
OBJECTIVE To prepare chitosan alginate microcapsule pulse preparation with polyelectrolyte layer-by-layer assembly and study its structure and in vitro release behavior. Methods The sodium alginate chitosan microgel core was prepared by using high voltage electrostatic capsule machine. The polyelectrolyte was coated on the surface of microgel by electrostatic adsorption principle to form the coating layer. The effects of different materials ratio on the swelling properties and release behavior of polyelectrolyte-coated microgels were investigated by single factor experiments. Results The results showed that the polyelectrolyte-coated microgels had a round appearance with a narrow particle size distribution. The in vitro release time lag was 2.67h. The drug release was rapid after lag time and reached 72% in 3h. Conclusions The microgel with polyelectrolyte as coating layer can be pulsed with sodium alginate as the core.
