目的检测乳腺癌组织中Notch1受体的表达水平,为乳腺癌的防治提供新的靶点。方法分别应用荧光定量PCR(FQ-PCR)和免疫组化技术检测60份乳腺癌组织中的Notch1受体蛋白及基因表达情况。实验设25份正常乳腺组织作对照。结果 FQ-PCR检测60份乳腺癌组织Notch1基因表达阳性,阳性率为87.5%,对照组为12.0%,DNA拷贝数分别为(3.66±0.37)×106拷贝/ml和(0.57±0.76)×102拷贝/ml,差异有统计学意义(P<0.05);免疫组织化学染色检测乳腺癌组织中Notch1分子表达阳性率为81.0%,正常乳腺组织阳性率为12.0%,差异有统计学意义(P<0.05)。结论乳腺癌组织高表达Notch1受体,表明Notch1受体与乳腺癌的发生、发展密切相关。
Objective To detect the expression of Notch1 receptor in breast cancer and provide a new target for the prevention and treatment of breast cancer. Methods The expression of Notch1 receptor and its gene in 60 breast cancer tissues were detected by fluorescence quantitative PCR (FQ-PCR) and immunohistochemistry respectively. Experimental set 25 normal breast tissue as a control. Results The positive expression rate of Notch1 gene in 60 breast cancer tissues was 87.5% by FQ-PCR and 12.0% in control group, respectively. The DNA copy number was (3.66 ± 0.37) × 106 copies / ml and (0.57 ± 0.76) × 102 (P <0.05). The positive rate of Notch1 expression in breast cancer was 81.0% and the positive rate of normal breast was 12.0% in immunohistochemical staining (P <0.05), the difference was statistically significant (P < 0.05). Conclusion High expression of Notch1 receptor in breast cancer indicates that Notch1 receptor is closely related to the occurrence and development of breast cancer.
