为了探讨壮腰健肾丸的缩尿作用及机制,该实验采用皮下注射150 mg·kg-1剂量的D-半乳糖诱导亚急性衰老模型,并将其分为模型组、缩泉丸组(1.17 g·kg-1·d-1)、壮腰健肾丸高、中、低剂量组(2.39,1.20,0.60 g·kg-1·d-1),另取正常动物设为空白组,连续给药8周。选用代谢笼法测量大鼠24 h尿量及水负荷5 h尿量;采用全自动生化仪检测尿中Na+,Cl-,K+浓度;应用ELISA法测定血清中醛固酮(aldosterone,ALD)、抗利尿激素(antidiuretic hormone,ADH)的含量;采用RT-PCR检测大鼠膀胱组织中P2X1与P2X3mRNA表达量的变化情况。结果显示壮腰健肾丸高、中剂量组对肾虚多尿大鼠24 h尿量及水负荷后5 h内尿量均显著下调(P<0.05,P<0.01);对尿中Na+和Cl-浓度均显著降低(P<0.01);血浆中ALD,ADH含量显著性升高(P<0.05,P<0.01);膀胱组织中P2X1与P2X3mRNA表达量显著下调(P<0.01);壮腰健肾丸低剂量组尿中Na+和Cl-浓度均显著降低(P<0.01)。结果说明壮腰健肾丸可能是通过下调肾虚多尿大鼠膀胱组织中P2X1与P2X3mRNA表达量从而发挥缩尿的作用。
In order to investigate the mechanism of oliguria of Zhuangyaojianshen Pills, subcutaneous injection of 150mg · kg-1 D-galactose was used to induce subacute aging model and divided into model group, 1.17 g · kg-1 · d-1), Zhuang Yao Jian Shen Wan high, medium and low dose groups (2.39,1.20,0.60 g · kg-1 · d-1), another normal animal as a blank group, Continuous administration for 8 weeks. Metabolic cage method was used to measure the urine volume of 24 hours and the water load of rats during 5 hours. The concentrations of Na +, Cl- and K + in urine were detected by automatic biochemical analyzer. The levels of aldosterone (ALD), antidiuresis (ADH). The expression of P2X1 and P2X3 mRNA in the bladder tissue of rats were detected by RT-PCR. The results showed that Zhuangyaojianshenwan high and medium dose groups had significantly decreased urine output at 24 h and urine volume within 5 h after water overload (P <0.05, P <0.01), while urinary Na + and Cl (P <0.01). The levels of ALD and ADH in plasma were significantly increased (P <0.05, P <0.01), while the expressions of P2X1 and P2X3 mRNA in bladder tissue were significantly decreased (P <0.01) Nephritis low dose group of urine Na + and Cl- concentrations were significantly lower (P <0.01). The results show Zhuangyaojianshen pill may play a role in reducing urine by down-regulating the expression of P2X1 and P2X3mRNA in the bladder tissue of rats with kidney deficiency and polyuria.
