目的:比较甘草内生菌与宿主甘草对痰浊阻肺模型大鼠的祛痰作用,筛选甘草内生菌有效菌株。方法:将Wistar大鼠70只随机分为7组,即正常对照组、模型组、复方贝母氯化铵阳性对照组、野生甘草水煎液组及甘草内生菌各组(JTYF023、JTYB006、JTYB029)。采用SO2烟薰、低温及冷风刺激诱导痰浊阻肺大鼠模型。造模10 d后,给予各组大鼠相应药物灌胃,连续7 d。进行证候学观察,检测酚红排泌量、肺功能指标4项、炎症相关因子(TNF-α、ICAM-1、COX-2)的含量、肺组织Na~+-K~+-ATP酶活性,并以免疫组化法测定水通道蛋白AQP1、AQP5的分布及表达,探讨药物的祛痰机制。结果:与正常对照组比较,模型组大鼠酚红排泌量增加,肺功能4项指标显著降低,炎症各相关因子的含量显著升高,Na~+-K~+-ATP酶活性及水通道蛋白AQP1、AQP5表达显著降低(P<0.01)。与模型组比较,水煎液组、JTYB029组酚红排泌量显著降低,肺功能4项指标显著升高,炎症各相关因子的含量显著降低,Na~+-K~+-ATP酶活性及水通道蛋白AQP1、AQP5表达显著升高(P<0.05或P<0.01)。结论:筛选出1株(JTYB029)与野生甘草水煎液祛痰作用相当的内生菌。
OBJECTIVE: To compare the expectorant effects of licorice endophyte and host licorice on phlegm-dampness-obstructing lung model rats and to screen effective strains of endophytic licorice. Methods: Seventy Wistar rats were randomly divided into seven groups: normal control group, model group, Fritillaria cirrhosa positive control group, wild licorice root decoction group and licorice endophyte group (JTYF023, JTYB006, JTYB029). Using SO2 smoke smoked, cold and cold wind-induced sputum turbid obstructive pulmonary disease model rats. After 10 days of modeling, the rats in each group were given the corresponding drugs for 7 days. Compared with the model group, the decoction group and the JTYB029 group significantly reduced the excretion of phenol red, the pulmonary function indicators were significantly increased, the levels of inflammation related factors were significantly decreased, Na ~ + -K ~ + -ATPase activity and The expression of AQP1 and AQP5 were significantly increased (P <0.05 or P <0.01). Conclusion: One strain of endophytic bacteria (JTYB029) which has the same expectorant effect as the wild liquorice decoction was screened out.
