目的探讨苦参碱对异体皮肤移植小鼠排斥反应的免疫抑制作用。方法 BALB/c小鼠作为供体,C57BL/6小鼠作为受体,建立背-背皮肤移植模型。术后分为假手术组、模型组、苦参碱组(20 mg/kg)、环孢素A(10 mg/kg)及联合组(苦参碱20 mg/kg+环孢素A 5 mg/kg),每组10只,皮下注射给药10 d。记录各只小鼠移植皮片的存活天数,术后第4天和第8天采用酶联免疫吸附试验检测血浆白介素-2(IL-2)含量。结果苦参碱组、环孢素A组、联合组小鼠移植皮片存活时间分别延长为(14.55±2.57)、(17.20±3.03)及(20.62±4.05)d,与模型组(11.69±1.84)d比较,组间差异均有统计学意义(P<0.01);联合组小鼠移植皮片存活时间比苦参碱组或环孢素A组均有显著延长(P<0.05或P<0.01);苦参碱组、环孢素A组及联合组第4天血浆IL-2水平分别为(4.37±1.24)、(2.51±0.92)及(3.19±0.86)ng/ml,与模型组(6.05±1.12)ng/ml比较,显著减少且组间差异均有统计学意义(P<0.01);苦参碱组、环孢素A组、联合组第8天血浆IL-2水平分别为(3.80±0.93)、(2.19±0.70)及(2.68±0.65)ng/ml,与模型组(6.94±2.06)ng/ml比较,显著减少且组间差异均有统计学意义(P<0.01)。结论苦参碱具有抑制异体皮肤移植小鼠免疫排斥反应的作用。
Objective To investigate the immunosuppressive effect of matrine on allogeneic skin graft rejection in mice. Methods BALB / c mice were used as donors and C57BL / 6 mice as recipients to establish a dorsal-dorsal skin graft model. The rats in sham operation group were divided into sham group, model group, matrine group (20 mg / kg), cyclosporine A (10 mg / kg) and combination group kg), 10 rats in each group, administered subcutaneously for 10 days. The survival days of transplanted skin graft in each mouse were recorded. The content of interleukin-2 (IL-2) was detected by enzyme linked immunosorbent assay on the 4th and 8th day after operation. Results The survival time of the transplanted skin in matrine group, cyclosporin A group and combined group mice were prolonged to (14.55 ± 2.57), (17.20 ± 3.03) and (20.62 ± 4.05) d, respectively, which were significantly higher than those in model group (11.69 ± 1.84 ) d, the difference between the two groups was statistically significant (P <0.01). The survival time of the transplanted skin graft in the combination group was significantly longer than that in the matrine group or cyclosporin A group (P <0.05 or P <0.01) (4.37 ± 1.24), (2.51 ± 0.92) and (3.19 ± 0.86) ng / ml respectively in the matrine group, cyclosporin A group and the combination group on the 4th day, 6.05 ± 1.12) ng / ml, the levels of IL-2 in matrine group, cyclosporine A group and combination group on the 8th day were respectively ((P <0.01) 3.80 ± 0.93, 2.19 ± 0.70 and 2.68 ± 0.65 ng / ml, respectively, which were significantly lower than those in model group (6.94 ± 2.06) ng / ml, with significant difference between the two groups (P <0.01). Conclusion Matrine inhibits immune rejection in allogeneic skin grafts in mice.
