本实验应用免疫组织化学染色 ,3H -TdR掺入 ,流式细胞仪检测技术探讨缺氧对大鼠肺动脉平滑肌细胞 (PSMC)增殖的影响。结果 :缺氧 12h其增殖细胞核抗原 (PCNA)阳性反应颗粒增加 ,2 4h明显增加 ,其含量是常氧组 2 .1倍。低氧 8h可使PSMC的3H -TdR摄取量减少 (P <0 .0 5 ) ,12h则促进其摄取 ,2 4h则明显较常氧对照组增多 (P <0 .0 5 )。流式细胞仪检测显示其G2 /M期细胞比例明显增多 (P <0 .0 1) ,G0 /G1 期细胞比例明显减少 (P <0 .0 1) ,细胞内总蛋白含量有所降低 (P <0 .0 1)。结论 :缺氧早期抑制或损害PSMC增殖 ,而后促使PSMC的表型改变 ,出现增殖 ,从一侧面证实缺氧可以直接促进PSMC增生。
In this study, immunohistochemical staining, 3H-TdR incorporation and flow cytometry were used to investigate the effect of hypoxia on the proliferation of pulmonary artery smooth muscle cells (PSMCs) in rats. RESULTS: After hypoxia for 12 hours, the number of PCNA positive reaction particles increased, increased significantly at 24 hours, and its content was 2.1 times that of normoxia group. Hypoxia for 8h reduced the uptake of 3H-TdR of PSMCs (P <0.05), uptake of PSMCs up to 12h, and increased uptake of PSMCs significantly (P <0.05) at 24 h. Flow cytometry showed that the proportion of cells in G2 / M phase increased significantly (P <0.01), the proportion of cells in G0 / G1 phase decreased significantly (P <0.01), the content of total protein in cells decreased P <0 .0 1). CONCLUSION: Hypoxia inhibits or inhibits the proliferation of PSMCs early, then causes the phenotype of PSMCs to change and proliferate. Hypoxic hypoxia can directly promote PSMC proliferation.
