目的探讨Krüppel样转录因子8(Krüppel-like factor 8,KLF8)和血管内皮生长因子(vascular endothelial growth factor,VEGF)蛋白在皮肤黑素瘤组织中的表达及其意义。方法30例原发性皮肤黑素瘤(primary malignant melanoma,PCMM)患者(PCMM组),13例转移性黑素瘤(metastatic malignant melanoma,MCMM)患者(MCMM组)和30例皮内痣患者(皮内痣组),3组采用免疫组织化学法检测不同组织中KLF8和VEGF的表达情况。结果 PCMM组和MCMM组KLF8和VEGF蛋白均呈强阳性表达,PCMM组和MCMM组KLF8和VEGF阳性率(73.33%、76.77%,76.92%、84.62%)和阳性细胞率((3.264±0.598)%、(5.382±0.839)%,(3.726±0.811)%、(5.534±1.376)%)均高于皮内痣组(13.33%、23.33%,(1.489±0.671)%、(2.750±0.592)%),差异有统计学意义(P<0.05);PCMM组与MCMM组比较差异无统计学意义(P>0.05);PCMM组和MCMM组KLF8与VEGF蛋白阳性率和阳性细胞率均呈正相关(r=0.912,P=0.000;r=0.859,P=0.006)。结论皮肤黑素瘤的发生、发展与KLF8和VEGF高表达有关。
Objective To investigate the expression of Krüppel-like factor 8 (KLF8) and vascular endothelial growth factor (VEGF) in cutaneous melanoma tissue and its significance. Methods Thirty patients with primary malignant melanoma (PCMM), 13 patients with MCMM (MCMM) and 30 patients with intradermal nevus Intradermal nevus group). Immunohistochemistry was used to detect the expression of KLF8 and VEGF in different tissues in 3 groups. Results The positive expression rates of KLF8 and VEGF in PCMM group and MCMM group were significantly higher than those in PCMM group and MCMM group (73.33%, 76.77%, 76.92%, 84.62%) and (3.264 ± 0.598)%, respectively , (5.382 ± 0.839)%, (3.726 ± 0.811)% and (5.534 ± 1.376)%, respectively, were significantly higher than those in the group of intradermal nevi (13.33%, 23.33%, (1.489 ± 0.671)%, (2.750 ± 0.592)%) (P <0.05). There was no significant difference between PCMM group and MCMM group (P> 0.05). The positive rates of KLF8 and VEGF protein in PCMM group and MCMM group were positively correlated with the positive cells (r = 0.912, P = 0.000; r = 0.859, P = 0.006). Conclusion The occurrence and development of cutaneous melanoma are related to the high expression of KLF8 and VEGF.
