目的:从体内外考察岗松总黄酮的抗炎作用。方法:采用脂多糖(LPS)刺激小鼠RAW264.7巨噬细胞模型,酶联免疫法(ELISA)检测岗松总黄酮对肿瘤坏死因子-α(TNF-α)及白介素-6(IL-6)的影响,蛋白印迹法(Western blot)检测细胞中诱导型一氧化氮合酶(i NOS)、环氧酶-2(COX-2)蛋白相对表达,考察其体外抗炎活性;采用小鼠耳廓肿胀、小鼠腹腔毛细血管通透性及大鼠棉球肉芽肿试验,考察其体内抗炎活性。结果:岗松总黄酮预处理能显著降低RAW264.7细胞中TNF-α及IL-6含量及i NOS、COX-2蛋白表达,并显著抑制二甲苯引起的小鼠耳廓肿胀、腹腔毛细血管通透性及大鼠棉球肉芽肿增生。结论:体内外试验均表明岗松总黄酮具有较好的抗炎活性,其机制可能与抑制炎性因子TNF-α、IL-6的释放及调控COX-2的表达有关。
OBJECTIVE: To investigate the anti-inflammatory effects of total flavonoids from Gangtong in vitro and in vivo. Methods: RAW 264.7 macrophages were stimulated with lipopolysaccharide (LPS). The levels of TNF-α and IL-6 were determined by enzyme-linked immunosorbent assay (ELISA) ), And the relative expression of iNOS and COX-2 protein were detected by Western blotting. The anti-inflammatory activity of iNOS and COX- Ear swelling, peritoneal capillary permeability in mice and rat granuloma test to investigate the anti-inflammatory activity in vivo. Results: Pretreatment with total flavonoids of P. xiaoensis could significantly decrease the content of TNF-α and IL-6, the expression of iNOS and COX-2 in RAW264.7 cells, and significantly inhibit the auricle swelling induced by xylene, Permeability and rat granuloma granuloma hyperplasia. CONCLUSION: Both in vitro and in vivo experiments showed that total flavonoids of P. xinggensis had good anti-inflammatory activity, which may be related to the inhibition of the release of inflammatory cytokines TNF-α and IL-6 and the regulation of COX-2 expression.
