目的:分离培养来源于人体的牙髓细胞并对其细胞表型进行分析。方法:采用组织块法培养人牙髓细胞,根据牙髓的细胞成分选择特异性抗体,利用免疫组化技术对其细胞表型进行分析。结果:III型胶原在体外培养的牙髓细胞中广泛表达,ON和OPN表达也较广泛,阳性细胞呈星形,伸出多个胞浆突起互相连接;α-平滑肌肌动蛋白和CD34表达细胞为集落状,前者呈细长的梭形,后者呈圆形;I型胶原、BSP和OC仅在少量细胞中表达,且细胞形态不规则;DSP、NF、CD14及CD45均为阴性。结论:体外培养的人牙髓细胞表型呈现多样性,表达平滑肌、内皮细胞及骨的标志物。
Objective: To isolate and culture human dental pulp cells and analyze their cell phenotypes. Methods: Human dental pulp cells were cultured by tissue block method. Specific antibodies were selected according to the cell components of dental pulp, and their cell phenotypes were analyzed by immunohistochemistry. Results: Type III collagen was expressed extensively in dental pulp cells cultured in vitro, and the expressions of ON and OPN were also more extensive. The positive cells were star-shaped and protruded from multiple cytoplasmic processes. The α-smooth muscle actin and CD34-expressing cells The former was elongated fusiform while the latter was circular. Type I collagen, BSP and OC were only expressed in a few cells, and the cells were irregular in shape. DSP, NF, CD14 and CD45 were all negative. CONCLUSION: The human dental pulp cells cultured in vitro present a diversity of phenotypes and express markers of smooth muscle, endothelial cells and bone.
