利用20个随机引物对厚皮甜瓜8个亲本与4个杂交种进行RAPD分析。10个引物的扩增DNA谱带具有多态性,亲本具有各自的特异谱带,绝大部分F1的扩增带型是双亲的互补带。各试材的相似系数范围为63.6%-96.5%。遗传距离聚类分析图表明,F1易与双亲或其一聚在一起。上述结果说明,可以利用RAPD标记在DNA水平上进行厚皮甜瓜的亲缘关系及纯度的鉴定。
RAPD analysis was performed on 8 parents and 4 hybrids of Muskmelon melon using 20 random primers. The amplified DNA bands of 10 primers were polymorphic, their parents had their own specific bands, and most of the amplification bands of F1 were the complementary bands of parents. The similarity coefficient of each test material ranged from 63.6% to 96.5%. Genetic distance cluster analysis shows that F1 is easy to get together with their parents or their parents. The above results show that RAPD markers can be used to identify the genetic relationship and the purity of Muskmelon at the DNA level.
