目的:探讨钙调蛋白在癌细胞增殖分化中的作用。方法:应用钙调蛋白拮抗剂—钙咪唑啉(CalmidazoliunCDZ)50~150nmol/L作用于人胃腺癌MGC803细胞,用白细胞计数法观察到CDZ对人胃癌细胞的增殖有明显的抑制作用,利用3HTdR掺入、PDE活性测定、CAMP含量测定、流式细胞光度术、cAMPPDEase电镜细胞化学技术进行系统研究。结果:钙调蛋白拮抗剂CDZ抑制MGC803细胞的DNA合成,一定浓度的CDZ阻断MGC803细胞于G1期,经CDZ处理的癌细胞内磷酸二脂酶(PhosphodiesterasePDE)活性下降,并使细胞内cAMP水平瞬时升高,癌细胞质膜上的3’.5’环磷酸腺苷磷酸二酯酶(cAMPPDEase)活性明显下降。结论:上述结果表明钙调蛋白在人胃癌细胞的增殖分化过程中起重要的调控作用,有理论和实际应用的价值。
Objective: To investigate the role of calmodulin in the proliferation and differentiation of cancer cells. Methods: The effect of CDZ on the proliferation of human gastric adenocarcinoma cell line MGC803 was induced by CalmodazoliunCDZ 50 ~ 150 nmol / L. The proliferation of human gastric cancer cell line CDZ was significantly inhibited by leukocyte counting. 3HTdR incorporation PDE activity determination, determination of CAMP content, flow cytometry, cAMPPDEase electron microscopy cytochemistry techniques for systematic study. Results: The CDZ of calmodulin inhibited the DNA synthesis of MGC803 cells. A certain concentration of CDZ blocked MGC803 cells in G1 phase. The activity of Phosphodiesterase PDE decreased in CDZ-treated cancer cells and the intracellular cAMP level Transiently increased, 3’.5 ’cAMPPDEase activity on the plasma membrane of cancer cells decreased significantly. Conclusion: These results indicate that calmodulin plays an important regulatory role in the proliferation and differentiation of human gastric cancer cells, and has theoretical and practical value.
