目的探讨壬基苯酚(NP)对H9c2心肌细胞内钙离子浓度和细胞增殖的影响以及GPR30受体的作用。方法采用细胞内钙测定方法来记录H9c2心肌细胞内钙离子浓度变化,采用MTT法来观察H9c2心肌细胞增殖程度。结果 1×10-10mol/L NP使细胞内钙离子浓度升高的幅度增加,并促进H9c2细胞增殖,而1×10-6mol/L NP使细胞内钙离子浓度升高的幅度降低,并抑制细胞增殖。G15可以阻断1×10-10mol/L NP对细胞内钙离子浓度和细胞增殖的促进作用,但不能阻断1×10-6mol/L NP对细胞内钙离子和细胞增殖的抑制作用。结论 NP对H9c2细胞的快速钙信号变化和细胞增殖具有非单调的剂量依赖性作用,其机制可能是由于GPR30受体在不同浓度下参与的机制不同。
Objective To investigate the effects of nonylphenol (NP) on intracellular calcium concentration and cell proliferation in H9c2 cardiomyocytes and the effect of GPR30 receptor. Methods Intracellular calcium measurement was used to record changes of intracellular calcium concentration in H9c2 cardiomyocytes. MTT assay was used to observe the proliferation of H9c2 cardiomyocytes. Results 1 × 10-10 mol / L NP increased the intracellular calcium concentration and promoted the proliferation of H9c2 cells. However, the concentration of 1 × 10-6 mol / L NP decreased the intracellular calcium concentration and inhibited Cell Proliferation. G15 could block the promotion of intracellular calcium concentration and cell proliferation by 1 × 10-10 mol / L NP, but could not block the inhibitory effect of 1 × 10-6 mol / L NP on intracellular calcium and cell proliferation. Conclusion NP has a non-monotonic dose-dependent effect on rapid calcium signaling and cell proliferation in H9c2 cells. The mechanism may be due to the different mechanism of GPR30 receptor participation at different concentrations.
