目的探讨ERα/ERβ、TGFβ1蛋白在乳腺癌发展过程中的表达变化。方法收集了2009年1月至2010年12月在山东大学第二医院乳腺外科及临沂市人民医院普外三科入院治疗的258例患者的病理资料,采用免疫组化方法检测正常乳腺组织(n=61)、乳头状瘤(n=65)、乳腺原位癌(n=45)及乳腺浸润性癌(n=87)组织中ERα/β、TGFβ1蛋白的表达情况。所有数据应用方差分析。结果 ERα/ERβ的比值在正常乳腺组织(0.34)、乳头状瘤(0.43)、原位癌(0.68)及浸润性癌细胞(0.71)呈逐步上升的趋势。TGFβ1总体表达情况在乳头状瘤、原位癌及浸润性癌与正常乳腺组织相比差异均有统计学意义(P<0.05),原位癌及浸润性癌强度分数高于乳管内乳头状瘤与正常乳腺组织(P<0.05)。结论在乳腺癌发生发展过程中ERα/ERβ升高,提示ERβ可能是乳腺癌的一个自我保护因素。TGF-β1蛋白表达可能是乳腺癌的早期事件,如能在早期阶段对TGFβ1的表达进行干预,可能对乳腺癌的防治起到重要作用。
Objective To investigate the expression changes of ERα / ERβ and TGFβ1 in the development of breast cancer. Methods From January 2009 to December 2010, pathological data of 258 patients admitted to Department of Breast Surgery, the Second Hospital of Shandong University and General Hospital of Linyi People’s Hospital were collected. Immunohistochemistry was used to detect normal breast tissue (n = 61), papilloma (n = 65), breast carcinoma in situ (n = 45) and invasive breast cancer (n = 87). All data were analyzed using ANOVA. Results The ratio of ERα / ERβ was gradually increased in normal breast tissues (0.34), papilloma (0.43), carcinoma in situ (0.68) and invasive cancer cells (0.71). The overall expression of TGFβ1 in papilloma, carcinoma in situ and invasive cancer compared with normal breast tissue were statistically significant (P <0.05), in situ carcinoma and invasive carcinoma intensity score higher than that of intraductal papilloma With normal breast tissue (P <0.05). Conclusion ERα / ERβ is elevated in the development of breast cancer, suggesting that ERβ may be a self-protective factor in breast cancer. TGF-β1 protein expression may be an early event in breast cancer. Being able to interfere with TGFβ1 expression at an early stage may play an important role in the prevention and treatment of breast cancer.
