目的探讨珍珠菜提取物(ZE4)诱导肿瘤细胞凋亡的作用。方法采用AO-EB双染法、Hoechst染色法观察ZE4给药后对人肝癌细胞株SMMC-7721肿瘤细胞形态学的影响;免疫荧光法检测ZE4给药后SMMC-7721细胞内凋亡蛋白及其配基(Fas/FasL)、肿瘤坏死因子相关凋亡诱导配体(TRAIL)、可溶性死亡受体5(DR5)、抑制凋亡蛋白(Bcl-2)及调控蛋白胱氨酸蛋白酶(Caspase3)的表达。结果Hochest染色和AO/EB双染结果表明,100μg/mlZE4可诱导SMMC-7721细胞发生凋亡,细胞凋亡的数目和形态改变随给药时间延长而变化明显;免疫荧光法检测结果表明,ZE4可通过上调Fas/FasL基因,上调TRAIL及DR5表达,下调Bcl-2表达,上调Caspase3表达等途径诱导细胞凋亡。结论珍珠菜提取物ZE4可诱导SMMC-7721肿瘤细胞发生凋亡,具有良好的抗肿瘤效果。
Objective To investigate the effect of ZE4 on tumor cell apoptosis. Methods AO-EB double staining and Hoechst staining were used to observe the effect of ZE4 on morphological changes of SMMC-7721 cells. Immunofluorescence method was used to detect the apoptosis proteins in SMMC-7721 cells after ZE4 administration The expressions of Fas / FasL, TRAIL, DR5, Bcl-2 and Caspase3 expression. Results The results of Hochest staining and AO / EB double staining showed that 100μg / ml ZE4 could induce the apoptosis of SMMC-7721 cells. The number of apoptotic cells and the morphological changes were obviously changed with the prolongation of administration time. The results of immunofluorescence showed that the expression of ZE4 It can induce apoptosis by up-regulating Fas / FasL gene, up-regulating TRAIL and DR5 expression, down-regulating Bcl-2 expression and up-regulating Caspase3 expression. Conclusion ZE4 extract can induce the apoptosis of SMMC-7721 tumor cells and has good anti-tumor effect.
