目的建立HPLC法测定木芙蓉提取物中金丝桃苷的方法,为木芙蓉提取物的开发利用提供科学依据。方法色谱柱为Diamonsil TM柱(250 mm×4.6 mm,5μm),流动相为乙腈-0.3%磷酸溶液(18∶82),体积流量1.0 mL/min,柱温30℃,检测波长360 nm。结果金丝桃苷与其他成分分离良好,在29.2~292.0 ng线性关系良好,r=0.999 9,平均回收率为98.7%,RSD为2.03%。结论所建立的定量方法简便、准确、专属性好、灵敏度高。
Objective To establish a HPLC method for the determination of Hibiscus extract from Hibiscus L. extract and provide a scientific basis for the exploitation and utilization of Hibiscus extract. Methods Diamonsil TM column (250 mm × 4.6 mm, 5 μm) was used. The mobile phase consisted of acetonitrile-0.3% phosphoric acid solution (18:82), the volume flow rate was 1.0 mL / min and the column temperature was 30 ℃. Results Hyperoside was well separated from other components and showed a good linear relationship between 29.2 and 292.0 ng (r = 0.999 9) with an average recovery of 98.7% and a RSD of 2.03%. Conclusion The established quantitative method is simple, accurate, specific and sensitive.
