CpG ODN佐剂序列IMB-AC5的体外免疫刺激活性分析

目的对CpG ODN候选佐剂IMB-AC5进行体外免疫刺激活性分析。方法以目前国外进入临床研究的两种CpG ODN佐剂序列(Coley 7909和Dynavax ISS1018)为对照,采用~3H-TDR法检测IMB-AC5刺激人外周血淋巴细胞(peripheral blood lymphocytes,PBLs)的增殖活性;流式细胞术分析CD19+B活化细胞表面CD69表达水平;ELISA法检测刺激后GEN细胞产生的IL-6和IFNα水平。结果 3种CpG ODN佐剂均能明显刺激人PBLs的增生,IMB-AC5刺激增生能力优于Dynavax ISS1018,略低于Coley 7909;IMB-AC5活化CD19+B细胞表面CD69表达水平与Coley7909相似,Dynavax ISS1018活化效果略低于前二者;3种CpG ODN佐剂体外活化树突状细胞水平差异较大,且呈明显的剂量依赖,IMB-AC5的最佳活化浓度范围为0.15~1.5μmol/L,Coley 7909的最佳活化浓度范围为<0.25μmol/L,活化效果均优于Dynavax ISS1018。结论 IMB-AC5可作为有效的疫苗候选佐剂,且不低于国外同类佐剂的体外免疫刺激活性。 Objective To investigate the immunostimulatory activity of CpG ODN candidate adjuvant IMB-AC5 in vitro. METHODS: Two kinds of CpG ODN adjuvant sequences (Coley 7909 and Dynavax ISS1018), which are currently in clinical trials abroad, were used as controls. The proliferation of peripheral blood lymphocytes (PBLs) stimulated by IMB-AC5 was detected by 3H-TDR assay The expression of CD69 on the surface of CD19 + B activated cells was analyzed by flow cytometry. The levels of IL-6 and IFNα produced by GEN cells were detected by ELISA. Results All three CpG ODN adjuvants could significantly stimulate the proliferation of human PBLs. The proliferation of IMB-AC5 was better than that of Dynavax ISS1018, which was slightly lower than that of Coley 7909. The expression of CD69 on IMB-AC5-activated CD19 + B cells was similar to that of Coley7909, The activation of ISS1018 was slightly lower than that of the former two. The dendritic cells activated by CpG ODN adjuvant differed significantly in a dose-dependent manner. The optimum activation concentration of IMB-AC5 was 0.15-1.5μmol / L , Coley 7909 the best activation concentration range <0.25μmol / L, the activation effect is better than Dynavax ISS1018. Conclusion IMB-AC5 can be used as an effective vaccine candidate adjuvant, and not less than the foreign similar adjuvants in vitro immunostimulatory activity.
