目的了解热疗联合丝裂霉素对大肠癌细胞的影响。方法本文用 MTT 法体外研究热疗联合丝裂霉素(MMC)对人大肠癌细胞株 LOVO 的细胞毒作用。结果单纯热,43℃ 30分钟具有较强的细胞毒作用。单纯化疗,在 MMC 浓度为0.01mg/ml 时,具有较强的细胞毒作用。热疗联合化疗,细胞毒作用明显增强,大于单纯热疗和单纯化疗之和,二者具有协同作用。43℃ 30分钟以上,细胞毒作用很强。单纯热疗和单纯化疗在低渗条件下,细胞毒作用大于等渗的细胞毒作用(P<0.05)。结论经尽可能短的时间,使温度达43℃,持续时间不少于30分钟,MMC 浓度不低于0.01mg/ml 时,热疗联合丝裂霉素对人大肠癌细胞有很强的细胞毒作用。
Objective To understand the effect of hyperthermia combined with mitomycin on colorectal cancer cells. Methods The cytotoxic effect of hyperthermia combined with mitomycin C (MMC) on human colorectal cancer cell line LOVO was studied in vitro using MTT assay. The results were simple and hot, 43 °C 30 minutes has a strong cytotoxic effect. Chemotherapy alone has a strong cytotoxic effect when the MMC concentration is 0.01 mg/ml. Hyperthermia combined with chemotherapy significantly increased cytotoxicity, which was greater than the sum of hyperthermia alone and chemotherapy alone. The two have synergistic effects. At 43°C for more than 30 minutes, the cytotoxicity is very strong. For hyperthermia and chemotherapy alone, cytotoxicity was greater than isotonic cytotoxicity in hypotonic conditions (P<0.05). Conclusion After the shortest possible time, the temperature reaches 43°C and the duration is not less than 30 minutes. When the MMC concentration is not less than 0.01 mg/ml, hyperthermia combined with mitomycin has strong cells on human colorectal cancer cells. Poisonous effect.
