目的观察中成药通心络对兔腹主动脉及骼动脉球囊损伤后血栓形成及内膜增生的影响。方法新西兰兔36只 ,随机分为3组 :通心络组16只 ,阿司匹林组11只 ,安慰剂组9只。各组服药剂量 :通心络0.38g·kg-1·d -1 ,阿司匹林50mg·kg-1·d -1,安慰剂组服用生理盐水10ml·d-1。1wk后制作腹主动脉和骼动脉球囊损伤模型 ,在模型制作后24h、1wk或1mon取受损节段血管 ,经HE染色后置显微镜下观察管腔内血栓形成及内膜增生情况。结果球囊损伤后24h、7d在各实验组均可见部分血管腹主动脉及髂动脉内膜增生 ,管腔狭窄5 %~40 % ,术后1mon安慰剂组2只兔腹主动脉及髂动脉内膜增生更为显著 ,管腔狭窄50%~70%。阿司匹林组血栓形成的血管比例少于另两组 ,但无显著差异。通心络组出现内膜增生的血管比例显著少于阿司匹林及安慰剂组。结论兔腹主动脉和髂动脉球囊损伤模型是研究PTCA术后再狭窄的理想模型。通心络明显抑制兔外周血管球囊损伤后内膜增生 ,提示通心络在再狭窄防治中可能有一定的潜在价值
Objective To observe the effect of Chinese traditional medicine Tongxinluo on thrombosis and intimal hyperplasia after balloon injury of rabbit abdominal aorta and iliac artery. Methods Thirty-six New Zealand rabbits were randomly divided into 3 groups: 16 in Tongxinluo group, 11 in aspirin group, and 9 in placebo group. The dosages of each group were: Tongxinluo 0.38g·kg-1·d -1 , aspirin 50mg·kg-1·d -1, and placebo group taking normal saline 10ml·d-1. The abdominal aorta and skeleton were made after 1wk In the arterial balloon injury model, the injured segmental vessels were harvested 24 hours, 1 week, or 1 month after the model was made. The thrombosis and intimal hyperplasia in the lumen were observed under a microscope after HE staining. RESULTS In 24h and 7d after balloon injury, the intimal hyperplasia of some vascular abdominal aorta and radial artery was seen in each experimental group. The lumen stenosis was 5% to 40%. After operation, 2 rabbits in the placebo group received abdominal aorta and radial artery. Intimal hyperplasia was more pronounced with a 50% to 70% narrowing of the lumen. The proportion of blood vessels in thrombosis in the aspirin group was less than that in the other two groups, but there was no significant difference. The proportion of vessels with intimal hyperplasia in the Tongxinluo group was significantly lower than that in the aspirin and placebo groups. Conclusion The rabbit abdominal aorta and radial artery balloon injury model is an ideal model for the study of restenosis after PTCA. Tongxinluo significantly inhibited intimal hyperplasia after balloon injury in rabbits, suggesting that Tongxinluo may have certain potential value in the prevention and treatment of restenosis.
