目的:探讨胃癌组织及其邻近胃组织中睾丸特异性Y样蛋白5(testis-specific Y-like protein5,TSPYL5)的表达对胃癌生物学行为的影响。方法:采用RT-PCR方法检测46例胃癌组织、癌旁胃正常粘膜组织中TSPYL5 mRNA的表达,分析其表达与胃癌生成、淋巴结转移、肿瘤浸润程度及分级的关系,同时采用MTT方法检测TSPYL5对胃癌细胞株AGS的影响。结果:与癌旁组织相比,46例胃癌组织中有26例TSPYL5 mRNA表达显著下调(56.5%),有淋巴结转移的胃癌组织中TSPYL5表达率与无淋巴结转移组织相比无显著差异(P>0.05),而在TNM中的Ⅱ、Ⅲ期以及不存在腹腔和远处转移时TSPYL5 mRNA表达显著下调(P<0.05)。同时TSPYL5对AGS细胞株生长有抑制作用。结论:TSPYL5 mRNA表达下调可能与胃癌的发生发展相关。
Objective: To investigate the effect of testis-specific Y-like protein 5 (TSPYL5) on the biological behavior of gastric cancer and its adjacent gastric tissues. Methods: The expression of TSPYL5 mRNA was detected by RT-PCR in 46 cases of gastric cancer and adjacent normal gastric mucosa. The relationship between TSPYL5 mRNA expression and gastric carcinogenesis, lymph node metastasis, tumor infiltration and grading was detected by MTT assay Effect of gastric cancer cell line AGS. Results: TSPYL5 mRNA was significantly down-regulated in 26 of 46 gastric cancer tissues (56.5%) compared with adjacent non-cancerous tissues. There was no significant difference in TSPYL5 expression between those with lymph node metastasis and those without lymph node metastasis (P> 0.05). However, the expression of TSPYL5 mRNA was significantly down-regulated in stage II and III of TNM and in the absence of peritoneal and distant metastasis (P <0.05). At the same time TSPYL5 AGS cell line growth inhibition. Conclusion: The down-regulation of TSPYL5 mRNA may be related to the occurrence and development of gastric cancer.
