目的观察健儿强身膏对哮喘豚鼠血清IL-10、IL-6、TNF-α表达的影响。方法采用卵蛋白(OVA)致敏法制备豚鼠哮喘模型,将48只豚鼠随机分为6组,即空白对照组、模型组、阳性对照组、健儿强身膏(高、中、低)3个剂量组,观察各组引喘潜伏期(T)及肺组织的病理变化,应用酶联免疫吸附试验法测定血清IL-10、IL-6、TNF-α的表达水平。结果各治疗组均可延长引喘潜伏期,与模型组相比差异有统计学意义(P<0.05)。与空白对照组相比,模型组的IL-6、IL-10表达显著增加(P<0.01),TNF-α的表达无显著差异;各治疗组与模型组相比IL-6、IL-10表达显著下降(P<0.01),TNF-α的表达无显著差异。结论健儿强身膏能下调哮喘豚鼠IL-6、IL-10的表达,可能为其抑制哮喘呼吸道炎症的重要作用机制之一。
Objective To observe the effect of Jian Er Qiang Gao Paste on the expression of IL-10, IL-6 and TNF-α in asthmatic guinea pigs. Methods The guinea pig asthma model was induced by ovalbumin (OVA) sensitization. 48 guinea pigs were randomly divided into 6 groups: blank control group, model group, positive control group, Jianliang Qiangtong Paste (high, medium and low) The latency of asthma and the pathological changes of lung tissue were observed. The levels of IL-10, IL-6 and TNF-α in serum were detected by enzyme linked immunosorbent assay (ELISA). Results All treatment groups could prolong the incubation period of asthma, which was significantly different from the model group (P <0.05). Compared with the blank control group, the expression of IL-6 and IL-10 in the model group was significantly increased (P <0.01), while the expression of TNF-α was no significant difference. Compared with the model group, IL-6 and IL- (P <0.01), while the expression of TNF-α had no significant difference. Conclusion Jian Er Qiang Gao can down-regulate the expression of IL-6 and IL-10 in asthmatic guinea pigs, which may be one of the important mechanisms of its inhibition of airway inflammation in asthma.
