1978~81年,作者对成都地区的机关干部和中小学生6479人进行了血红蛋白病的普查,发现了几种异常血红蛋白(Hb)并进行了化学结构分析,现报道如下. 方法先取末梢血,用简快Hb电泳法(1)筛选,有异常Hb区带者进行体检和家族调查,并取静脉血作血红蛋白测定和红细胞形态观察,Hb的醋酸纤维素薄膜和淀粉胶电泳,HbA_2和异常Hb定量,抗硷Hb测定,不稳定性试验(异丙醇试验、热变性试验、变性珠蛋白小体检查),Hb溶解度测定及异常Hb肽链分析(8M尿素解离和醋纤膜肽链电泳法):然后作异常Hb的化学结构分析,即先将Hb溶液制成珠蛋白干粉,经CM—22纤维素柱
