目的通过测定并比较柱果铁线莲、单叶铁线莲、威灵仙、山木通、毛蕊铁线莲、转子莲、女萎和天台铁线莲的ITS序列,为铁线莲属药用植物的分子鉴定提供依据。方法利用PCR法从叶片基因组DNA中扩增ITS,借助Clustal X、MEGA、DNASTAR等软件比较和分析ITS的序列特征。结果铁线莲属8种药用植物ITS1与ITS2之间的长度为534~561 bp,变异位点50个,信息位点22个;天台铁线莲与转子莲的遗传距离最小,与女萎遗传距离最大,亲缘关系最远。序列已提交至NCBI数据库,登录号为JF714638-JF714645。结论获得铁线莲属8种药用植物的ITS序列,为分子鉴定奠定了基础。
Objective To determine and compare the ITS sequences of Clematis, Clematis, Clematis, Samutpratus, Mint, Clematis, Wildebeest and Tail of Clematis, Use plant molecular identification provide the basis. Methods ITS was amplified by PCR from genomic DNA of leaves. The sequences of ITS were compared and analyzed by Clustal X, MEGA and DNASTAR softwares. Results The lengths of ITS1 and ITS2 among 8 species of medicinal plants in Clematis ranged from 534 to 561 bp, with 50 loci and 22 loci. The genetic distance between Clematis tricuspidata and Drosophila melanogaster was the lowest, The largest genetic distance, the farthest genetic relationship. The sequence has been submitted to the NCBI database under accession number JF714638-JF714645. Conclusion The ITS sequences of eight medicinal plants of Clematis were established, which laid the foundation for molecular identification.
