目的:优化青藤碱脂质体的膜材处方,并进行制备工艺研究。方法:采用乙醚注入法制备脂质体,以包封率为指标,采用混料均匀设计法优化其膜材处方,确定豆磷脂、胆固醇、维生素E等组分的用量。并在此基础上采用单因素考察法进行制备工艺研究,包括缓冲液用量、制备温度、超声时间等。结果:最佳膜材处方为青藤碱:豆磷脂:胆固醇:维生素E质量化(8.92:60.35:28.81:1.91),缓冲液用量为50 mL.g-1膜材,制备温度为50℃,超声时间为20 min。结论:按此优化处方及工艺制备的脂质体可获得满意的形态及包封率。
Objective: To optimize the membrane prescription of sinomenine liposome and to study its preparation process. Methods: The liposomes were prepared by ether injection method. The encapsulation efficiency was taken as the index. The formulation of the membrane was optimized by the uniform design of mixture. The dosage of soybean lecithin, cholesterol and vitamin E were determined. And on this basis, the use of single factor method to study the preparation process, including the amount of buffer, preparation temperature, ultrasonic time and so on. Results: The optimal prescription of sinomenine was: soybean lecithin: cholesterol: the quality of vitamin E (8.92: 60.35: 28.81: 1.91), the amount of buffer was 50 mL.g-1, the preparation temperature was 50 ℃, Ultrasound time was 20 min. Conclusion: According to this optimized formulation and process of preparation of liposomes can obtain satisfactory morphology and encapsulation efficiency.
