目的探讨TRPV1和TRPV2基因表达水平与北京地区儿童哮喘的关系。方法实时定量PCR方法检测38例健康儿童,46例哮喘儿童外周血淋巴细胞中TRPV1和TRPV2 mRNA表达水平差异。免疫印迹法定量检测总IgE水平,尘螨、春季花粉、秋季花粉、蟑螂和猫狗毛、真菌等特异性IgE水平,对检测结果进行多因素logistic回归分析。结果健康和哮喘组儿童TRPV1和TRPV2的mRNA表达水平差异有统计学意义(P<0.01),真菌特异性IgE表达水平差异有统计学意义(P<0.01)。结论 TRPV1和TRPV2基因表达水平升高可能是北京地区儿童哮喘发病的危险因素。
Objective To investigate the relationship between TRPV1 and TRPV2 gene expression and childhood asthma in Beijing. Methods The expression of TRPV1 and TRPV2 mRNA in peripheral blood lymphocytes of 38 healthy children and 46 asthmatic children were detected by real-time PCR. Quantitative IgE levels of total IgE, dust mites, spring pollen, autumn pollen, cockroaches and cats, dogs and cats, and fungi were detected by Western blotting. Multivariate logistic regression analysis was performed on the test results. Results The mRNA expressions of TRPV1 and TRPV2 in healthy and asthmatic children were significantly different (P <0.01), while the fungal specific IgE expression was significantly different (P <0.01). Conclusion The increased expression of TRPV1 and TRPV2 may be the risk factors of childhood asthma in Beijing.
