PZ6菌株是一株分离于香蕉根际土壤的具有生防潜力的木霉菌株,通过rDNA-ITS鉴定为棘孢木霉Trichoderma asperellum。室内测定不同条件下菌丝生长、产孢量,研究其生物学特性。结果表明,PZ6菌株在PDA、PSA、OMA和Czapek培养基上均能生长,PDA和OMA培养基上菌丝生长和产孢最佳。在15~37℃均能生长,25~30℃菌丝生长较好,以30℃菌丝生长最快,培养3d菌落直径达到89.00mm,且产孢量也显著高于其他培养温度。在pH值2~12的范围均可以生长和产孢,最适生长产孢为pH值5~9。葡萄糖、果糖、乳糖、麦芽糖和蔗糖都能够较好促进菌丝生长,葡萄糖和果糖是PZ6菌株产孢的最佳碳源;硫酸盐与硝酸盐是PZ6菌丝生长和产孢的最佳氮源。光照对菌丝生长影响明显,持续光照可显著促进产孢。
The PZ6 strain is a Trichoderma strain with biocontrol potential isolated from banana rhizosphere soil and identified as Trichoderma asperellum by rDNA-ITS. Indoor determination of mycelial growth, sporulation under different conditions, to study the biological characteristics. The results showed that the PZ6 strain could grow on the PDA, PSA, OMA and Czapek medium, the mycelial growth and sporulation on the PDA and OMA medium were the best. The growth of mycelia at 25 ~ 30 ℃ was the fastest, the mycelial growth at 30 ℃ was the fastest, the diameter of colonies was 89.00mm, and the sporulation amount was also significantly higher than other culture temperature. Growth and sporulation can be carried out in the pH range of 2 ~ 12, and sporulation in the optimum pH range of 5 ~ 9. Glucose, fructose, lactose, maltose and sucrose all could promote mycelial growth better. Glucose and fructose were the best carbon sources for sporulation of PZ6 strain. Sulfate and nitrate were the best nitrogen source for PZ6 mycelial growth and sporulation . Light on the growth of mycelium obvious, continuous light can significantly promote sporulation.
