以橙黄色果肉西瓜‘WM-Clr-2’及红色果肉西瓜‘PI 179881’为试材,采用HPLC法分析其番茄红素及β–胡萝卜素在7个发育时期的积累情况,同时采用q RT-PCR技术分析番茄红素和β–胡萝卜素代谢途径中8个关键酶基因(PSY1、PSY2、PDS、ZDS、CRTISO、LCYB、NCED1和NCED7)的表达量。结果显示,随着果实的发育,番茄红素和β–胡萝卜素含量均不断增加,且在同一时期,红色果肉‘PI 179881’中的含量始终大于‘WM-Clr-2’。q RT-PCR结果显示PSY1在‘PI 179881’中的表达量高于‘WM-Clr-2’,LCYB和NCED1在‘WM-Clr-2’中的表达量高于红色果肉‘PI 179881’。分析番茄红素及β–胡萝卜素的积累规律与相关基因表达规律说明PSY1的大幅上调表达,LCYB的下调表达及NCED1的低表达可能是红色果肉‘PI 179881’中番茄红素和β–胡萝卜素大量积累的主要原因。PSY1的低表达水平,LCYB和NCED1的高表达水平可能是橙黄色果肉‘WM-Clr-2’中番茄红素及β–胡萝卜素积累量少的主要原因。
The accumulation of lycopene and β-carotene in seven developmental stages was analyzed by HPLC with orange-yellow fruit watermelon ’WM-Clr-2’ and red fruit watermelon ’PI 179881’ The expression levels of eight key enzyme genes (PSY1, PSY2, PDS, ZDS, CRTISO, LCYB, NCED1 and NCED7) in lycopene and β-carotene metabolic pathways were analyzed by PCR-PCR. The results showed that the content of lycopene and β-carotene increased with the development of fruit, and the contents of ’PI 179881’ were always higher than that of ’WM-Clr-2’ in the same period. q RT-PCR results showed that the expression level of PSY1 in ’PI 179881’ was higher than that of ’WM-Clr-2’, and that of LCYB and NCED1 in ’WM-Clr-2’ was higher than that of ’PI 179881’. Analysis of the law of accumulation of lycopene and β-carotene and the related gene expression pattern indicated that the expression of PSY1 was significantly up-regulated. The down-regulated expression of LCYB and the low expression of NCED1 might be the expression of lycopene and β-carotene in red pulp ’PI 179881’ The main reason for a large number of accumulation. The low expression level of PSY1 and the high expression levels of LCYB and NCED1 may be the main reason for the low accumulation of lycopene and β-carotene in orange-yellow pulp ’WM-Clr-2’.
