目的对石家庄市集中空调系统水中分离的25株嗜肺军团菌进行脉冲场凝胶电泳(PFGE)分析,初步建立石家庄市嗜肺军团菌的PFGE分型数据库。方法采用PFGE技术,对2008年石家庄市集中空调系统水分离的嗜肺军团菌用AscI酶切,将PFGE图像用BioNumerics 4.0软件处理后,采用统一的分子量标准对照沙门菌(Braenderup)H9812菌株进行校准后进行聚类分析。结果 25株嗜肺军团菌的PFGE图谱共分为13种不同的PFGE带型,其中,LPA16.SJZ0006(7株)带型最多,占分离菌株的28.0%。聚类分析结果显示,所有血清1型的菌株间相似性系数为60%~100%;血清2~14型的菌株间相似性系数为80%~100%。结论本次从石家庄市集中空调系统水分离的嗜肺军团菌菌株呈多态性分布,基因组变异较大,同时也具有克隆化特征。
Objective To analyze the PFGE of 25 Legionella pneumophila isolated from water of centralized air-conditioning system in Shijiazhuang and to establish the PFGE typing database of Legionella pneumophila in Shijiazhuang. Methods PFGE was used to isolate Legionella pneumophila isolated from water of central air-conditioning system in Shijiazhuang in 2008. After the PFGE images were processed with BioNumerics 4.0 software, a uniform molecular weight standard was used to calibrate the strain of Brahman’s H9812 After clustering analysis. Results The PFGE patterns of 25 Legionella pneumophila strains were divided into 13 different PFGE patterns. Among them, the band patterns of LPA16.SJZ0006 (7 strains) were the largest, accounting for 28.0% of the isolates. Cluster analysis showed that the similarity coefficient of all serotype 1 isolates was between 60% and 100%, and the similarity coefficient of serotypes 2 to 14 was between 80% and 100%. Conclusion The strains of Legionella pneumophila isolated from water of central air-conditioning system in Shijiazhuang City are polymorphic, with large variation of genome and clonal characteristics.
