自认识HbA_2测定对诊断杂合子型β-地中海贫血的重要性之后20年,尚无一个精确测定HbA_2的方法被普遍接受。作者介绍一个简单的测定HbA_2方法。材料:醋酸纤维薄膜条,5.7×14cm。不连续缓冲系统:(1)电泳槽缓冲剂-0.058M,pH 8.6巴比妥缓冲剂(巴比妥钠10.3g,巴比妥1.4g,加蒸馏水至1,000ml)。(2)浸膜缓冲剂——tris-EDTA-硼酸缓冲剂(TEB),0.12M,pH9.1(tris12.1g EDTA-Na_21.56g,硼酸0.92g,加蒸馏水至1,000ml)。膜溶剂——80%醋酸。
Since the recognition of the importance of HbA 2 in the diagnosis of heterozygous β-thalassemia 20 years later, no precise method for the determination of HbA 2 has been universally accepted. The authors introduce a simple method for the determination of HbA_2. Material: acetate film strip, 5.7 × 14cm. Discontinuous buffer system: (1) electrophoresis tank buffer -0.058M, pH 8.6 barbiturate buffer (sodium butyrate 10.3g, barbiturate 1.4g, add distilled water to 1,000ml). (2) Leaching Buffer - Tris-EDTA-Borate Buffer (TEB), 0.12 M, pH 9.1 (tris 12.1 g EDTA-Na-21.56 g, boric acid 0.92 g, plus distilled water to 1,000 ml). Membrane solvent - 80% acetic acid.
