目的:龙胆中龙胆苦苷的分离制备和含量测定。方法:采用HPLC测定龙胆中龙胆苦苷的含量和HSCCC分离制备龙胆苦苷;HPLC条件:以甲醇、0.1%磷酸溶液(甲醇:0.1%磷酸溶液=24:76)为流动相;流速1m L/min;检测波长270nm;进样量10μL,柱温25o C。结果:经测定,龙胆草中龙胆苦苷的含量为0.5411%。选出正丁醇-水(1:1)为最佳溶剂体系,固定相保留率36.0%,分配系数0.714,上相作固定相,下相作流动相,转速为850r/min,主机正转,流速2m L/min,分离制备出的龙胆苦苷经HPLC检测,纯度可达98%以上。结论:HSCCC仪器性能可靠、分析成本低、易于操作,可分离制备出高纯度的龙胆苦苷。
Objective: Isolation, preparation and determination of gentiopicroside in gentian. Methods: The determination of gentiopicroside in gentian and the isolation of gentiopicroside by HSCCC were carried out by HPLC. The HPLC conditions were methanol and 0.1% phosphoric acid solution (methanol: 0.1% phosphoric acid solution = 24: 76) 1m L / min; detection wavelength 270nm; injection volume 10μL, column temperature 25o C. Results: The content of gentiopicroside in gentian was 0.5411%. The optimal solvent system was n-butanol-water (1: 1). The retention rate of stationary phase was 36.0%. The partition coefficient was 0.714. The upper phase was the stationary phase and the lower phase was the mobile phase at 850 r / min. , Flow rate 2m L / min, isolated and prepared gentiopicroside by HPLC, the purity of up to 98%. Conclusion: The HSCCC instrument has reliable performance, low cost, easy operation and high purity gentiopicroside.
