本文研究IL2和IL-2受体在人T细胞增殖中的作用.方法是从健康人外周血分离出巨噬细胞和T细胞.巨噬细胞经γ-射线照射并用神经氨酸酶和半乳糖氧化酶(NAGO)处理作为刺激细胞.T细胞与经上述方法处理的自身巨噬细胞共同培养,经不同时间终止激活.然后通过E花环再次分离T细胞予以重新培养,培养中加或不加外源性IL-2,用~3H-TdR掺入法检测其增殖反应.T细胞与巨噬细胞共同培养至少经2-4小时激活,可产生对IL-2的反应,而以激活后18-24小时反应最强.用抗IL-2受体的单
This article investigates the role of IL2 and IL-2 receptors in the proliferation of human T cells by separating macrophages and T cells from healthy human peripheral blood. The macrophages are irradiated with gamma-rays and treated with neuraminidase and galactose Oxidase (NAGO) was used as stimulator cells.T cells were co-cultured with autologous macrophages treated as described above, and then activated at different times.Afterwards, T cells were isolated and re-cultured by E-rings, with or without Proliferation response of IL-2 was detected by ~ 3H-TdR incorporation method.Culturing T cells and macrophages for at least 2-4 hours could produce response to IL-2, 24 hours the strongest response with a single anti-IL-2 receptor
