雷竹笋汁对糖尿病大鼠血浆多种因子和肝糖原的影响(英文)

背景:糖尿病患者糖代谢紊乱与脂代谢异常互为因果,导致肝糖原减少和血浆多种因子改变,从而易致心、肾、脑等血管并发症的发生,寻找有效的糖尿病二级干预药物已成为当今研究的热门课题。目的:探讨雷竹笋汁对大鼠肝糖原、血浆葡萄糖及血脂的干预效应。设计:以实验动物为研究对象,随机对照观察研究。单位:咸宁学院药学院实验室。材料:Wistar雄性大鼠40只,体质量(230±20)g,由广州军区武汉总医院医学实验动物中心提供。方法:实验于2002-09/2003-06在咸宁学院药学院实验室完成采用四氧嘧啶(200mg/kg)腹腔注射制备糖尿病大鼠模型。分别用不同剂量的雷竹笋汁犤205.70mg/kg(雷竹笋-Ⅰ组)和436.40mg/kg(雷竹笋-Ⅱ组)犦灌胃28d,并以健康大鼠作正常对照。测定各组大鼠肝糖原含量和血浆多种因子水平。主要观察指标:各组大鼠肝糖原、血浆葡萄糖、三酰甘油、总胆固醇、非酯化脂肪酸(non-esterifiedfattyacid,NEFA)、低密度脂蛋白胆固醇(Lowdensitylipoproteincholesterol,LDL-c)、高密度脂蛋白胆固醇(highdensitylipoproteincholesterol,HDL-c)、丙二醛含量和超氧化物歧化酶(superoxidedismutase,SOD)活性。结果:雷竹笋-Ⅱ组大鼠血浆葡萄糖含量为(9.65±1.14)mmol/L、三酰甘油为(1.31±0.18)mmol/L、NEFA为(0.51±0.04)mmol/L、LDL-c为(0.79±0.19)mmol/L和丙二醛为(6.31±0.83)nmol/L显著低于模型组犤葡萄糖(16.14±1.59)mmol/L,三酰甘油(1.94±0.31)mmol/L,NEFA(0.59±0.05)mmol/L,LDL-c(1.34±0.28)mmol/L和丙二醛(10.95±2.90)nmol/L,(t=3.478~9.379,P<0.01)犦,而雷竹笋-Ⅱ组HDL-c为(1.09±0.13)mmol/L、SOD为(39.76±6.23)NU/ml和肝糖原为(6.70±0.62)mg/g显著高于模型组犤HDL-c(0.55±0.11)mmol/L,SOD(23.24±3.35)NU/ml和肝糖原(4.06±1.24)mg/g,(t=5.388~9.000,P<0.01)犦。而且,雷竹笋汁二个剂量组间LDL-c的降低作用和SOD及肝糖原的升高作用呈现剂量依赖关系(P<0.05~0.01)。结论:雷竹笋汁明显降低糖尿病大鼠血浆葡萄糖、三酰甘油、NEFA、LDL-c和MDA及升高HDL-c、SOD和肝糖原等因子水平,显示有效的抗糖尿病作用。 Background: Diabetes mellitus patients with glucose metabolism disorders and abnormal lipid metabolism are mutual cause and effect, leading to decreased liver glycogen and a variety of plasma factor changes, which easily lead to heart, kidney, brain and other vascular complications, to find effective drugs for the secondary treatment of diabetes Has become a hot topic in today’s research. OBJECTIVE: To investigate the effect of bamboo shoot juice on liver glycogen, plasma glucose and blood lipid in rats. Design: Randomized controlled observational study of experimental animals. Unit: Xianning College School of Pharmacy. MATERIALS: Forty Wistar male rats, weighing (230 ± 20) g, were provided by the Medical Laboratory Animal Center of Wuhan General Hospital of Guangzhou Military Command. METHODS: The experimental diabetic rat model was prepared by intraperitoneal injection of alloxan (200 mg/kg) in the laboratory of the College of Pharmacy of Xianning University from September 2002 to October 2003. Different doses of Lei bamboo shoot juice 犤 205.70mg/kg (Leizhusun-I group) and 436.40mg/kg (Leizhusun-II group) were given intragastrically for 28 days and healthy rats were used as normal controls. The liver glycogen content and plasma levels of various factors were determined in each group. MAIN OUTCOME MEASURES: Rat liver glycogen, plasma glucose, triacylglycerol, total cholesterol, non-esterified fatty acid (NEFA), low density lipoprotein cholesterol (LDL-c), high-density lipids High-density lipoprotein cholesterol (HDL-c), malondialdehyde content and superoxide dismutase (SOD) activity. RESULTS: The plasma glucose levels in the Lei bamboo shoot-II group were (9.65±1.14) mmol/L, triacylglycerol (1.31±0.18) mmol/L, NEFA (0.51±0.04) mmol/L, and LDL-c. (0.79±0.19)mmol/L and malondialdehyde (6.31±0.83) nmol/L were significantly lower than those in the model group (16.14±1.59) mmol/L, triacylglycerols (1.94±0.31) mmol/L, NEFA (0.59±0.05) mmol/L, LDL-c (1.34±0.28) mmol/L and malondialdehyde (10.95±2.90) nmol/L, (t=3.478 to 9.379, P<0.01) 犦, and Lei Zhusun - In Group II, HDL-c was (1.09±0.13) mmol/L, SOD was (39.76±6.23) NU/ml, and glycogen was (6.70±0.62) mg/g, which was significantly higher than that of model group 犤HDL-c (0.55±). 0.11 mmol/L, SOD (23.24±3.35) NU/ml and hepatic glycogen (4.06±1.24) mg/g, (t=5.388 to 9.000, P<0.01). Moreover, the decrease of LDL-c and the increase of SOD and hepatic glycogen in a dose-dependent manner (P<0.05-0.01) in two dose groups of bamboo shoot juice. Conclusion: Leizhusun juice significantly reduced plasma glucose, triglyceride, NEFA, LDL-c and MDA, and increased HDL-c, SOD and hepatic glycogen levels in diabetic rats, indicating an effective anti-diabetic effect.
