[目的]观察鱼藤素与氟尿嘧啶联用对胃癌细胞的作用。[方法]胃癌MGC-803细胞经鱼藤素、氟尿嘧啶单药及两药联合处理48h后,MTT法检测其对细胞生长增殖抑制水平,采用等效图解法分析其作用;流式细胞术检测经鱼藤素处理后MGC-803细胞周期的变化。[结果]鱼藤素可剂量依赖性抑制MGC-803细胞增殖,有G1期阻滞作用,与氟尿嘧啶联用可明显增强细胞杀伤作用。[结论]鱼藤素与氟尿嘧啶联用可产生明显协同作用,机制为两者作用于不同细胞周期,增强了细胞对氟尿嘧啶的敏感性。
[Objective] To observe the effect of deguelin and fluorouracil on gastric cancer cells. [Methods] The gastric cancer MGC-803 cells were treated with deguelin and fluorouracil monotherapy and the two drugs for 48 hours. MTT assay was used to detect the inhibitory effect on the proliferation of MGC-803 cells. Equivalent graphical method was used to analyze the effect of MGC-803 cells. Flow cytometry Changes of cell cycle of MGC-803 treated with deguelin. [Result] The results showed that deguelin could inhibit the proliferation of MGC-803 cells in a dose-dependent manner, with the effect of blocking G1 phase. Combined with fluorouracil, deguelin could obviously enhance the cytotoxicity. [Conclusion] The combination of deguelin and fluorouracil can produce obvious synergistic effect. The mechanism is that the two act on different cell cycle and enhance the sensitivity of cells to fluorouracil.
