目的观察大肠癌细胞上EGF_R与EGF_Au结合后定位、内吞、循环和代谢过程。方法应用透射电镜观察胶体金标记的表皮生长因子(Au_EGF)与HT_29细胞上EGF_R的作用过程。结果发现加入Au_EGF数分钟就结合于细胞膜上EGF_R,10min内金颗粒主要储存中于细胞膜,10_20min主要散布于细胞浆,细胞核内也有少量金颗粒存在,30_50min胞浆中金颗粒逐渐减少消失,小部分金颗粒重又返回细胞膜。完成一个循环约需30_50min,但细胞核内金颗粒至120min时仍不消失。结论EGF与EGF_R结合后可发生典型的受体内吞和循环再生过程。
Objective To observe the localization, endocytosis, circulation and metabolism of EGF_R and EGF_Au on colorectal cancer cells. Methods Transmission electron microscopy was used to observe the interaction between colloidal gold-labeled epidermal growth factor (Au_EGF) and EGF_R on HT_29 cells. The results showed that the Au_EGF was bound to the EGF_R on the cell membrane within several minutes. The gold particles were mainly stored in the cell membrane within 10 min, scattered mainly in the cytoplasm in 10~20 min, and there was also a small amount of gold particles in the cell nucleus. The gold particles in the cytoplasm gradually decreased and disappeared within 30~50 min. The gold particles return to the cell membrane again. It takes about 30-50 minutes to complete a cycle, but the gold particles in the nucleus do not disappear until 120 minutes. Conclusion The typical receptor endocytosis and recycling process can occur after the combination of EGF and EGF_R.
