本文利用杂交瘤技术获得SZ-21单克隆抗体,属IgGl亚类。以亲和层析法纯化Sz-21抗体所识别的抗原,SDS-PAGE表明其分子量在未还原时为14万和9.2万,还原后为13万和11万,PAS染色阳性,因而是血小板膜糖蛋白Ⅱb/Ⅲa复合物。转移电泳技术进一步表明,SZ-21与分子量为9.2万的蛋白结合,证明它特异作用于血小板膜糖蛋白Ⅲa(GP Ⅲa)。以0.2mg/ml糜蛋白酶处理血小板30分钟后,SZ-21所识别的抗原决定簇仍存在于血小板膜上,转移电泳方法表明
In this paper, hybridoma technology to obtain SZ-21 monoclonal antibody, IgGl subclass. The antigens recognized by the Sz-21 antibody were purified by affinity chromatography. SDS-PAGE showed that the molecular weight of the Sz-21 antibody was 140,000 and 92,000 without reduction and 130,000 and 110,000 after reduction, respectively. PAS staining was positive and thus platelet membrane Glycoprotein Ⅱ b / Ⅲ a complex. Transient electrophoresis further demonstrated that SZ-21 binds to a protein of 92,000 in molecular weight, demonstrating its specific effect on platelet membrane glycoprotein IIIa (GPIIIa). After treating platelets with 0.2 mg / ml chymotrypsin for 30 minutes, the antigenic determinants identified by SZ-21 are still present on the platelet membrane and the method of transfer electrophoresis
