目的:探讨金属蛋白酶-2/金属蛋白酶组织抑制因子2、非活化金属蛋白酶-2前酶/金属蛋白酶组织抑制因子2比值与肝细胞癌侵袭力的关系。方法:利用十二烷基硫酸钠-聚丙烯酰胺凝胶电泳酶谱和反向酶谱分析技术对32例肝细胞癌组织中金属蛋白酶-2和金属蛋白酶组织抑制因子2含量及非活化的金属蛋白酶-2前酶/金属蛋白酶组织抑制因子-2,金属蛋白酶-2/金属蛋白酶组织抑制因子2比值进行检测并对比分析。结果:金属蛋白酶组织抑制因子2的分子量为18 000;癌旁组织中非活化金属蛋白酶-2的前酶/金属蛋白酶组织抑制因子2(0.94),金属蛋白酶-2/金属蛋白酶组织抑制因子2(0.42)均<1.0;肝细胞肝癌组织中非活化金属蛋白酶-2前酶/金属蛋白酶组织抑制因子2(1.70),金属蛋白酶-2/金属蛋白酶组织抑制因子2(1.64)均>1.0;转移组的2项比值也明显高于无转移组(P<0.01)。结论:非活化金属蛋白酶-2前酶/金属蛋白酶组织抑制因子2,金属蛋白酶-2/金属蛋白酶组织抑制因子2的异常变化可能是评价肝细胞癌生物学行为的有力指标。
Objective: To investigate the relationship between MMP-2 / TIMP-2, MMP-2 / TIMP-2 and invasiveness of hepatocellular carcinoma. Methods: The expressions of tissue inhibitor of metalloproteinase-2 and metalloproteinase-2 in 32 cases of hepatocellular carcinoma (HCC) were determined by sodium dodecyl sulfate-polyacrylamide gel electrophoresis and reversed-phase zymography. Protease-2 preenzyme / metalloproteinase-2, metalloproteinase-2 / metalloproteinase 2 ratio were detected and compared. Results: The molecular weight of tissue inhibitor of metalloproteinase 2 was 18 000. The expression of MMP-2 / MMP-2 (0.94), MMP-2 / TIMP-2 0.42) were all less than 1.0. The levels of non-active metalloproteinase-2proteinase / metalloproteinase 2 (1.70) and metalloproteinase-2 / metalloproteinase 2 (1.64) The two ratios were also significantly higher than those without metastasis (P <0.01). Conclusion: Abnormal changes of non-active MMP-2 / TIMP-2, TIMP-2 / MMP-2 may be useful indicators for evaluating the biological behavior of hepatocellular carcinoma.
