目的:观察基质金属蛋白酶-2(MMP-2)、基质金属蛋白酶-9(MMP-9)和IV型胶原(IV-C)在糖尿病大鼠肾组织中的表达及灵芝的干预作用。方法:40只雄性SD大鼠随机分为正常对照组(C组)、糖尿病未治疗组(DM组)、糖尿病灵芝(1.2g·kg-1·d-1)治疗组(GL组),用免疫组化方法(SP法)检测治疗后各组大鼠肾皮质MMP-2、MMP-9及IV-C的表达,同时进行光镜、透射电镜观察各组大鼠肾皮质病理改变。结果:DM组大鼠肾小球MMP-2、MMP-9蛋白表达较C组明显降低,IV-C蛋白表达明显升高,出现糖尿病肾病典型的病理改变。GL组MMP-2、MMP-9蛋白较DM组表达明显升高,IV-C表达较DM组明显降低,病理改变好转。结论:灵芝通过上调MMP-2、MMP-9蛋白表达,减少细胞外基质IV-C积聚而对糖尿病大鼠起到肾脏保护作用。
Objective: To observe the expression of matrix metalloproteinase-2 (MMP-2), matrix metalloproteinase-9 (MMP-9) and type IV collagen (IV-C) in the kidney of diabetic rats and the intervention of Ganoderma lucidum. METHODS: Forty male Sprague-Dawley rats were randomly divided into normal control group (C group), diabetic untreated group (DM group) and diabetic ganoderma lucidum (1.2 g·kg-1·d-1) treatment group (GL group). The expression of MMP-2, MMP-9, and IV-C in renal cortex of rats in each group was detected by immunohistochemical method (SP method). At the same time, pathological changes of renal cortex were observed by light microscope and transmission electron microscope. RESULTS: The expression of MMP-2 and MMP-9 protein in glomeruli of DM rats was significantly lower than that in C group, and the expression of IV-C protein was significantly increased. The pathological changes of diabetic nephropathy were typical. The expression of MMP-2 and MMP-9 in GL group was significantly higher than that in DM group. The expression of IV-C was significantly lower than that in DM group, and the pathological changes were improved. CONCLUSION: Ganoderma lucidum protects diabetic rats by up-regulating the expression of MMP-2 and MMP-9 protein and reducing the accumulation of extracellular matrix IV-C.
