目的观察黄芪对糖尿病肾病(DN)的作用。方法以STZ诱导糖尿病动物模型,采用免疫组化和RTPCR检测方法观察黄芪对糖尿病大鼠肾皮质TGFβ蛋白、基因表达的影响。结果糖尿病大鼠肾皮质TGFβ免疫染色强度比未治疗大鼠明显减轻,TGFβmRNA水平比未治疗大鼠明显下降(P<0.01),黄芪与开搏通合用可增强上述作用。结论黄芪可抑制糖尿病大鼠肾皮质TGFβ的过度表达,其防治DN发生、发展的作用机制可能部分是通过抑制TGFβ的过度表达。
Objective To observe the effect of Astragalus on diabetic nephropathy (DN). Methods STZ-induced diabetic animal models were used to observe the effect of astragalus on renal cortex TGFβprotein and gene expression in diabetic rats by immunohistochemistry and RTPCR. Results The intensity of TGFβ immunoreactivity in renal cortex of diabetic rats was significantly lower than that of untreated rats. The level of TGFβ mRNA was significantly lower than that of untreated rats (P<0.01). The combination of jaundice and captopril enhanced the above effects. Conclusion Astragalus can inhibit the overexpression of TGF-β in the renal cortex of diabetic rats. The mechanism of its action may be partly through inhibiting the overexpression of TGFβ.
