目的检测硒蛋白K(Sel K)在小鼠脾、肾、肠、肺、脑、肝组织中的表达差异及在T淋巴细胞内的定位,为研究Sel K的分布与生物学功能的关联性提供参考。方法采用实时荧光定量PCR,Western blot以及免疫组化方法检测硒蛋白K在小鼠各组织的表达差异,并采用细胞免疫化学的方法 ,用激光共聚焦显微镜观察硒蛋白K在小鼠T淋巴细胞内的定位。结果实时荧光定量PCR结果显示硒蛋白K在小鼠各组织中均有表达,并且在脾脏中的表达明显高于其他组织(P<0.05)。Western blot及免疫组化结果与实时荧光定量PCR结果基本一致。细胞免疫化学结果显示硒蛋K在小鼠T淋巴细胞中定位于内质网。结论硒蛋白K是一种在脾脏中高表达的内质网蛋白。
Objective To detect the expression of selenoprotein K (Sel K) in spleen, kidney, intestine, lung, brain and liver of mice and its localization in T lymphocytes. To study the relationship between the distribution of Sel K and the biological function for reference. Methods The expression of selenoprotein K in various tissues of mice was detected by real-time fluorescence quantitative PCR, Western blot and immunohistochemistry. The expression of selenoprotein K was detected by laser confocal microscopy in mouse T lymphocytes Within the positioning. Results The results of real-time quantitative PCR showed that selenoprotein K was expressed in all tissues of mice and was significantly higher in spleen than in other tissues (P <0.05). Western blot and immunohistochemistry results were consistent with real-time fluorescence quantitative PCR results. Immunocytochemical results showed that selenium K was located in the endoplasmic reticulum in mouse T lymphocytes. Conclusion Selenoprotein K is a kind of endoplasmic reticulum protein highly expressed in spleen.
