目的:探讨五味子冻干粉改善果蝇睡眠作用的量效与时效关系。方法:在恒温、恒湿、自动光控条件下,以7日龄果蝇作为观察对象,分别给以含药浓度为0.25%,0.5%,1%,2%,4%,8%的培养基(每只培养指管分别含冻干粉30,60,120,240,480,960 mg),利用果蝇活动监测仪(DAMS),以静止时间持续5 min以上作为睡眠标准,观察并分析24 h内睡眠总时间的变化,探讨量效关系;在此基础上以不同时长给药,观察其24 h内睡眠总时间变化,探讨时效关系。结果:雌果蝇在8%药物培养基、给药6 d情况下睡眠延长最明显;而雄果蝇在8%药物培养基、给药4 d情况下睡眠延长最明显。结论:五味子冻干粉可以改善果蝇睡眠,并在一定范围内呈现量效和时效关系。
OBJECTIVE: To investigate the relationship between the dose-effect and the time-effect of Fructus Schisandrae freeze-dried powder to improve the sleep of Drosophila melanogaster. Methods: Drosophila melanogaster at 7 days old under the conditions of constant temperature, constant humidity and automatic light control were used to observe the effects of different concentrations of Drosophila melanogaster on the culture medium containing 0.25%, 0.5%, 1%, 2%, 4%, 8% (30, 60, 120, 240, 480 and 960 mg of lyophilized powder in each culture tube). Drosophila activity monitor (DAMS) was used to measure the total sleep time within 24 h , To explore the dose-effect relationship; on this basis, with different duration of administration, observed within 24 h of total sleep time changes, to explore the relationship between the aging. Results: In the 8% drug medium, female flies had the most prolonged sleep prolongation after 6 d administration, while the male flies most prolonged sleep in the 8% drug medium for 4 d. Conclusion: Fructus Schisandrae freeze-dried powder can improve the sleep of Drosophila melanogaster, and show dose-effect and time-effect relationship within a certain range.
