目的:探讨孕早期高中链脂肪酸膳食(HMCFAD)对子代肥胖和肥胖相关基因肉毒碱棕榈酰基转移酶1(CPT1)表达的影响,寻找早期防治肥胖的靶标及合理的孕期膳食。方法:Wistar大鼠孕早期分别食用标准膳食(SD)和HMC-FAD,所产雄性子鼠分别为对照(CON)组和高中链脂肪酸(HMF)组,断乳后食用SD至成年;将CON组大鼠随机分为两组:CON组(食用SD)和高脂对照(HFC)组。HFC和HMF组大鼠高脂膳食诱导肥胖。分期处死大鼠取材,Real-Time PCR法检测骨骼肌中CPT1 mRNA的表达。结果:与CON和HFC组相比,HMF组大鼠的体重降低(P<0.05);CPT1表达持续升高。结论:孕早期HMCFAD可程序性升高子代CPT1基因的表达,降低成年肥胖的发生。
Objective: To investigate the effect of early pregnancy high-middle-chain fatty acid diet (HMCFAD) on the expression of obesity-related gene carnitine palmitoyltransferase 1 (CPT1) in the offspring and to find out the target of early prevention and treatment of obesity and reasonable diet during pregnancy. Methods: Wistar rats were fed with standard diet (SD) and HMC-FAD in the first trimester of pregnancy. The male mice were divided into control group (CON) and high-middle-chain fatty acid group (HMF) Rats were randomly divided into two groups: CON (SD) and HFC (high fat control) groups. High fat diet induces obesity in HFC and HMF rats. The rats were sacrificed by staging and Real-Time PCR was used to detect the expression of CPT1 mRNA in skeletal muscle. Results: Compared with CON and HFC groups, the weight of rats in HMF group decreased (P <0.05), and CPT1 expression continued to increase. Conclusion: HMCFAD can increase the expression of CPT1 gene in offspring progesterone in early pregnancy, and reduce the incidence of adult obesity.
