目的探讨何首乌含药血清体外对骨髓间充质干细胞(MSCs)增殖的影响及机理研究。方法分离、培养大鼠MSCs,何首乌含药血清诱导培养MSCs 72h后,MTT法检测大鼠MSCs增殖情况;RT-PCR检测增殖过程中不同细胞因子mRNA表达;Real-time PCR检测SCF mRNA表达;Western blot检测SCF蛋白表达;ELISA法检测培养上清液SCF含量。结果 10%何首乌含药血清具有明显促进MSCs增殖的作用,并明显促进SCF mRNA表达及可溶性SCF蛋白的分泌。结论 10%何首乌含药血清具有促进MSCs增殖的作用,可能与其促进SCF mRNA表达、诱导MSCs分泌可溶性SCF蛋白增多有关。
Objective To investigate the effects of Polygonum multiflorum containing drug serum on the proliferation of bone marrow mesenchymal stem cells (MSCs) and its mechanism. Methods The MSCs were isolated and cultured, the MSCs were cultured for 72 hours, and the proliferation of MSCs was detected by MTT assay. The mRNA expression of different cytokines was detected by RT-PCR. The expression of SCF mRNA was detected by Real-time PCR. The SCF protein expression was detected by Western blot and the SCF content in culture supernatant was detected by ELISA. Results 10% Polygonum multiflorum containing serum significantly promote the proliferation of MSCs, and significantly promote SCF mRNA expression and soluble protein secretion of SCF. Conclusion 10% Polygonum multiflorum containing serum can promote the proliferation of MSCs, which may be related to its promotion of SCF mRNA expression and induction of increased secretion of soluble SCF protein by MSCs.
