目的:探讨Fbxw7在肝癌中的表达及其与肝癌细胞增殖能力的相关性。方法:收集40例肝细胞癌组织及对应的癌旁组织,运用RT-PCR、免疫组化技术检测Fbxw7在肝癌及对应癌旁组织中的表达情况;real time RT-PCR技术检测正常肝细胞株LO2、肝癌细胞株Hep3B和SMMC-7721中Fbxw7 mRNA的表达水平;平板克隆形成实验与裸鼠皮下成瘤实验检测不同Fbxw7 mRNA表达水平的细胞株体内外增殖能力。结果:Fbxw7 mRNA及蛋白在肝癌组织中表达水平显著低于对应癌旁组织(P<0.05);Fbxw7蛋白低表达与高Edmonson分级和高TNM分期具有显著的相关性(P<0.05);正常肝细胞株LO2中Fbxw7 mRNA的表达水平显著高于肝癌细胞株Hep3B和SMMC-7721(P<0.05);Fbxw7表达较低的细胞株形成的克隆数较多,且裸鼠皮下成瘤体积较大,结果均具有统计学差异(P<0.05)。结论:Fbxw7低表达与肝癌的恶性临床病理特征相关,并与肝癌细胞增殖能力相关。
Objective: To investigate the expression of Fbxw7 in hepatocellular carcinoma and its correlation with the proliferation of hepatoma cells. Methods: Forty hepatocellular carcinoma tissues and their corresponding paracancerous tissues were collected. RT-PCR and immunohistochemistry were used to detect the expression of Fbxw7 in hepatocellular carcinoma (HCC) and corresponding paracancerous tissues. Real time RT-PCR was used to detect the expression of Fbxw7 in normal hepatocellular carcinoma LO2, Fbxw7 mRNA expression in Hep3B and SMMC-7721 cells. The proliferation of cell lines with different Fbxw7 mRNA expression levels in vitro were detected by plate clone formation assay and subcutaneous tumor formation assay in nude mice. Results: The expression of Fbxw7 mRNA and protein in hepatocellular carcinoma was significantly lower than that in paracancerous tissues (P <0.05). The expression of Fbxw7 protein was significantly correlated with high Edmonson grade and high TNM stage (P <0.05) The expression level of Fbxw7 mRNA in LO2 cell line was significantly higher than that in Hep3B and SMMC-7721 cells (P <0.05). The cell lines with lower Fbxw7 expression formed more colonies, The results were statistically significant (P <0.05). Conclusion: The low expression of Fbxw7 is associated with the clinicopathological features of hepatocellular carcinoma and its correlation with the proliferation of hepatoma cells.
