目的探讨胃癌生长过程中胃周淋巴管、小血管分布及变化规律。方法以SGC- 7901胃癌组织块植入裸鼠胃壁,尔后每隔1周处死1批,取胃周组织切片、双酶染色和苏木素-伊红(HE)染色,光镜观察淋巴管及微血管并图像分析,荷瘤组为实验组,与正常裸鼠对照。结果早期荷瘤鼠的淋巴管及微小血管面积、直径、不规则程度、密度与对照组比较无明显变化(P>0.05);随肿瘤生长,胃周淋巴管面积、最大直径、平均直径呈总体增加趋势,植瘤3周后较3周前比较差异有统计学意义(P<0.05),总体荷瘤鼠淋巴管密度比正常裸鼠高,差异有统计学意义(P<0.05);胃周血管面积、最大直径、平均直径、不规则程度随肿瘤生长呈总体增高趋势,植瘤2周后较2周前比较差异有统计学意义(P<0.05),密度变化差异无统计学意义(P>0.05);原位植瘤6周后可见胃周5’-核苷酸酶(5’-Nase)活性管道与碱性磷酸酶(ALPase)活性管道存在相通现象。结论随肿瘤生长胃周组织淋巴管密度增加、管径增粗;血管变化以管径增粗为主。晚期移植瘤的胃周血管可能与淋巴管存在相通现象。
Objective To investigate the distribution and changes of gastric lymphatic vessels and small vessels in the process of gastric cancer growth. Methods The stomach of nude mice was implanted with SGC-7901 gastric cancer tissue and sacrificed one week every other week. The sections of the stomach were stained by double enzyme staining and HE staining. The lymphatic vessels and microvessels were observed with light microscope Image analysis, tumor-bearing group for the experimental group, and normal control. Results There was no significant difference in the area, diameter, irregularity and density of lymphatics and microvessels in early tumor-bearing mice compared with the control group (P> 0.05). With the growth of the tumor, the lymphatic area, the maximum diameter and the average diameter (P <0.05). The density of lymphatics in the whole tumor-bearing mice was higher than that in normal nude mice (P <0.05), and the difference was statistically significant Vascular area, maximum diameter, average diameter and irregularity increased with tumor growth. There was a significant difference between two weeks after tumor implantation (P <0.05) and no significant difference in density (P > 0.05). After 6 weeks of in situ implantation, the 5’- Nase activity in the stomach could be found in the same way as the activity of alkaline phosphatase (ALPase). Conclusion With the increase of lymphatic vessel density in gastric peritumoral tissue, the diameter of the vessel increases. The change of blood vessel is dominated by the increase of diameter. Late gastric transplanted tumor may be related to the presence of lymphatic vessels and the phenomenon.
