以中华红叶杨茎段为外植体,研究了温度及光照条件对其褐变的影响。结果表明:外植体低温预处理与简单培养基培养相结合,抑制褐化效果好;培养初期进行暗培养能有效降低褐化率,以暗培养10d为宜;温度对褐化率也有影响,在一定范围内,温度越高,褐化越严重,低温可较好地减轻褐化;光照强度1 000~3 000lx对褐化率的影响不大。
In order to study the effects of temperature and light conditions on the browning of stem segments, The results showed that the low temperature pretreatment of explants combined with the simple culture medium inhibited the browning effect; dark culture in the early culture could effectively reduce the browning rate, which was suitable for dark culture for 10 days; the temperature also had an influence on the browning rate, In a certain range, the higher the temperature, the more severe the browning and the lower the browning temperature. The light intensity 1000 ~ 3000lx had little effect on the browning rate.
