目的研究高糖对大鼠施万细胞(SCs)生长及凋亡的影响,并探讨其可能机制。方法将培养的SCs分别用含不同浓度葡萄糖的培养液孵育72h,四甲基偶氮唑盐(MTT)检测细胞活力,原位缺口末端标记法(TUNEL)检测细胞凋亡,Western blotting检测Bcl-2及Bax的表达。结果高浓度葡萄糖(60mmol/L、90mmol/L)组SCs活力降低,细胞凋亡率增加,抗凋亡蛋白Bcl-2表达下降及凋亡蛋白Bax表达升高。结论高糖抑制SCs生长,诱导细胞凋亡,可能与Bcl-2表达下降及Bax表达升高有关。
Objective To investigate the effects of high glucose on the growth and apoptosis of rat Schwann cells (SCs) and to explore its possible mechanism. Methods Cultured SCs were incubated with different concentrations of glucose for 72 hours respectively. MTT assay was used to detect cell viability. Apoptosis was detected by TUNEL. Western blotting was used to detect Bcl- 2 and Bax expression. Results The viability of SCs in high concentration glucose (60 mmol / L, 90 mmol / L) group was decreased, the apoptosis rate was increased, the expression of anti-apoptotic protein Bcl-2 and the apoptosis protein Bax were increased. Conclusion High glucose can inhibit the growth of SCs and induce apoptosis, which may be related to the decrease of Bcl-2 expression and the increase of Bax expression.
