目的:探讨雌、孕激素对分泌中期子宫内膜腺上皮细胞高峰表达上皮型钙粘附蛋白(E-cadherin)的调节。是否与一氧化氮(NO)及白介素-1(IL-1)有关。方法:取人分泌中期子宫内膜,分离腺上皮,原代培养24小时,分别加入一氧化氮合酶抑制剂L-NAME、白介素-1受体拮抗剂IL-Ira,以PBS对照,继续培养24小时后,用间接免疫荧光法标记腺上皮细胞E-cadhefin抗原,流式细胞仪测定标记阳性率及平均荧光强度。结果:抑制NO产生或阻断IL-1r均使分泌期子宫内膜腺上皮细胞表达E-cadherin降低。结论:NO或IL-1可能是雌、孕激素调节子宫内膜表达E-cadherin的途径之一。
Objective: To investigate the regulation of estrogen and progesterone on the peak expression of E-cadherin in secretory endometrial glandular epithelial cells. Whether with nitric oxide (NO) and interleukin-1 (IL-1) related. Methods: The human endomembrane was secreted and the glandular epithelium was isolated. Primary culture was performed for 24 hours. Nitric oxide synthase inhibitor L-NAME and interleukin-1 receptor antagonist IL-Ira were added respectively. Twenty-four hours later, the E-cadhefin antigen of glandular epithelial cells was labeled by indirect immunofluorescence. The positive rate and the average fluorescence intensity of the marker were determined by flow cytometry. Results: Inhibition of NO production or blocking of IL-1r decreased E-cadherin expression in secretory endometrial glandular epithelial cells. Conclusion: NO or IL-1 may be one of the pathways by which estrogen and progesterone regulate the expression of E-cadherin in endometrium.
