目的探讨EphB4在前列腺癌(PCa)组织中的表达及其与淋巴管生成和肿瘤转移的关系。方法应用免疫组织化学法检测40例PCa和10例良性前列腺增生(BPH)组织中EphB4和LYVE-1蛋白的表达,根据LYVE-1染色结果计数淋巴管密度(LVD)。结果相比BPH,EphB4在PCa组织中高表达,LVD值低于BPH;在激素非依赖性前列腺癌(AIPC)组织中EphB4表达阳性率显著高于激素依赖性前列腺癌(ADPC)组织;肿瘤为中低分化、临床病理分期>T2、有淋巴结转移及远处转移的PCa组织中EphB4阳性率、LVD值显著高于肿瘤高分化、临床病理分期≤T2、无淋巴结转移及远处转移的癌组织,其差异均具有统计学意义(P<0.05);即EphB4表达、LVD值与PCa的肿瘤分化程度呈负相关,与肿瘤临床病理分期、淋巴结转移及远处转移呈正相关。结论 EphB4在PCa的发生发展和PCa的淋巴管生成中起重要作用,其促进肿瘤浸润和转移。阻断EphB4抑制PCa淋巴管生成,可能成为PCa治疗的新靶点。
Objective To investigate the expression of EphB4 in prostate cancer (PCa) and its relationship with lymphangiogenesis and tumor metastasis. Methods The expressions of EphB4 and LYVE-1 protein in 40 cases of PCa and 10 cases of benign prostatic hyperplasia (BPH) were detected by immunohistochemistry. LVD was counted according to LYVE-1 staining. Results Compared with BPH, EphB4 was highly expressed in PCa tissues and the LVD value was lower than that in BPH. The positive expression of EphB4 in hormone-independent prostate cancer (AIPC) was significantly higher than that in hormone-dependent prostate cancer (ADPC) The poorly differentiated, clinical pathological stage> T2, the positive rate of EphB4 in PCa tissues with lymph node metastasis and distant metastasis was significantly higher than that in well-differentiated tumor with clinical pathological stage ≤ T2, without lymph node metastasis and distant metastasis, The difference was statistically significant (P <0.05). EphB4 expression and LVD were negatively correlated with the degree of differentiation of PCa tumor and positively correlated with the clinicopathologic stage, lymph node metastasis and distant metastasis. Conclusion EphB4 plays an important role in the development of PCa and lymphangiogenesis in PCa, which promotes tumor invasion and metastasis. Blocking EphB4 inhibits PCa lymphangiogenesis may be a new target for PCa therapy.
