目的研究活血解毒方对糖尿病大鼠神经病变的预防作用,探讨其作用机制。方法观察活血解毒方对糖尿病大鼠周围神经病变的疗效及作用机理。雄性SD大鼠分为正常组、模型组、活血解毒方组。链脲佐菌素(streptozocin,STZ)腹腔注射60 mg/kg造模,观察大鼠血糖、热板实验潜伏期、甩尾时间、坐骨神经病理的改变,检测血清及坐骨神经中抑制羟自由基、超氧阴离子能力和过氧化氢、丙二醛(malondialdehyde,MDA)含量。结果模型组大鼠血糖显著升高,热板潜伏期延长,甩尾时间延长(P<0.01),坐骨神经结构显著改变;活血解毒方组热板潜伏期及甩尾时间显著缩短(P<0.01),坐骨神经病理明显改善。模型组大鼠血清中抑制羟自由基及超氧阴离子能力显著降低(P<0.01),活血解毒方组血清中抑制羟自由基及超氧阴离子能力升高(P<0.05或P<0.01),血清及坐骨神经中MDA含量下降(P<0.05)。结论活血解毒方可能对糖尿病大鼠神经病变有预防作用,其机制可能与抑制氧化应激有关。
Objective To study the preventive effect of Huoxue Jiedu Fang on neuropathy in diabetic rats and to explore its mechanism. Methods To observe the curative effect and mechanism of Huoxue Jiedu Fang on peripheral neuropathy in diabetic rats. Male SD rats were divided into normal group, model group, Huoxue Jiedu Fang group. Rats were injected intraperitoneally with streptozocin (STZ) at a dose of 60 mg / kg to observe the changes of blood glucose, hot plate incubation time, flick latency and sciatic nerve pathology. Serum and sciatic nerve inhibition of hydroxyl radical, Anion capacity and hydrogen peroxide, malondialdehyde (MDA) content. Results The blood glucose of rats in model group was significantly higher than that in control group (P <0.01). The latency of the hot plate was longer than that of the control group (P <0.01), and the sciatic nerve structure was significantly changed. Pathology improved significantly. The ability of inhibiting the hydroxyl radical and superoxide anion in serum of model group was significantly lower than that of model group (P <0.01), and the ability of inhibiting the free radical and superoxide anion in serum of Huoxue Jiedu Fang group was increased (P <0.05 or P <0.01) Serum and sciatic nerve MDA content decreased (P <0.05). Conclusions Huoxue Jiedu Fang may prevent neuropathy in diabetic rats, which may be related to the inhibition of oxidative stress.
