目的探讨促排卵药对小鼠子宫内膜发育的影响,为临床用药提供实验依据。方法随机将160只雌性昆明小鼠分为非孕组和受孕组,每组又分为对照组、克罗米酚组(CC组)、单纯促排卵组(HMG+HCG组)和GnRHa+单纯促排卵组(GnRHa+HMG+HCG组)4个小组,每小组20只。各用药组分别给予相应的促排卵药,对照组给予生理盐水。于用药后24 h和48 h分批处死非孕组小鼠,取子宫做成石蜡切片,HE染色,观察子宫内膜发育情况。受孕组的分组和用药与非孕组完全相同,于非孕组雌鼠处死取材的当天与雄鼠合笼,让雌鼠受孕。于受孕后6.5 d处死孕鼠,观察孕鼠着床情况以及胚胎发育指标。结果非孕组:对照组子宫内膜厚度明显厚于3个用药组(P<0.05),3个用药组中,GnRHa+单纯促排卵组子宫内膜厚度以及腺体发育略优于另外两组,但无显著性差异(P>0.05)。受孕组:对照组胚胎着床率明显高于用药组(P<0.05),各用药组之间有差异,但无统计学意义(P>0.05)。结论在三种促排卵方案中,GnRHa+单纯促排卵方案对子宫内膜的影响最小,内膜与间质发育的同步性最好,着床率较高。
Objective To investigate the effects of ovulation induction drugs on the development of mouse endometrium and to provide experimental evidence for clinical use. Methods 160 female Kunming mice were randomly divided into non-pregnant group and conception group, and each group was divided into control group, clomiphene phenol group (CC group), simple ovulation induction group (HMG + HCG group) and GnRHa + simplex Ovulation group (GnRHa + HMG + HCG group) 4 groups, each group of 20. Each treatment group were given the corresponding ovulation drugs, the control group given saline. Non-pregnant mice were sacrificed in batches at 24 h and 48 h after treatment, paraffin sections of the uterus were made and HE staining was performed to observe the development of the endometrium. Pregnancy group grouping and medication and non-pregnant group exactly the same, in the non-pregnant group of female rats were sacrificed on the day of males and cages, so that female rats conceive. The pregnant rats were killed at 6.5 days after conception, and the implantation status and embryo development of pregnant rats were observed. Results In non-pregnant group, the thickness of endometrium in control group was significantly thicker than that in control group (P <0.05). The thickness of endometrium and glandular development in GnRHa + simple ovulation group were better than the other two groups in three groups But no significant difference (P> 0.05). In the control group, the embryo implantation rate in the control group was significantly higher than that in the control group (P <0.05). There was a significant difference between the two groups (P> 0.05). Conclusion Among the three ovulation induction programs, GnRHa + simple ovulation induction program has the least effect on the endometrium, the synchronicity of intima-media development and implantation rate are higher.
